Research Specialist II The Dalton Laboratory is seeking a highly motivated Research Specialist II to conduct research into the biologic and therapeutic implications of spliceosome mutations in myeloid malignancies (acute myeloid leukemia and myelodysplastic syndromes). The laboratory uses gene editing to engineer and then study unique human cell models of cancer-associated mutations in spliceosome genes and validates findings in primary human patient samples. Biological studies include analysis of mutation-induced RNA splicing, proteome changes, and diverse cellular pathway alterations including those in energy metabolism. Therapeutic studies include targeting metabolic weaknesses, characterizing therapeutic antibody targets, and developing novel immunotherapeutic modalities against spliceosome-mutant cells. The Dalton Laboratory exists in the State-of-the-Art Bunting Blaustein Cancer Research Building and is part of the Hematologic Malignancies Department at the Sidney Kimmel Johns Hopkins Comprehensive Cancer Center. This position requires experience in laboratory techniques in molecular and cellular biology. Specific Duties & Responsibilities: Develops experimental designs and has responsibility for planning, implementation, and interpretation results; design process may be in collaboration with the PI or completed independently. Identifies problems, troubleshoots and analyzes variations observed and reported in regular testing protocols; modifies, refines or adapts techniques and procedures; modifies and/or adjusts quality control measures. Coordinates and/or conducts multiple, concurrent assignments. Compiles, analyzes, and interprets research data. Monitors documentation of results; reviews and recognizes documentation, which may lead to modification and adaptation of research methodologies; may collaborate with others; Adapts and/or modifies experimental methods based upon interpretation of data. Participates in editing and preparing manuscripts, scientific illustrations, and computer graphics for publication. Oversees and assures laboratory compliance with regulatory requirements and JHU standards, policies and procedures. In conjunction with other laboratory staff and the PI, helps to keep inventory and order laboratory supplies and equipment. Minimum Qualifications (Mandatory): BS/BA in Biological Sciences, Chemistry or related field required Master's Degree in Biotechnology, preferred. Minimum of 2 years' laboratory experience performing relevant techniques in a comparable setting. Master's Degree, with related graduate research, may substitute for experience to the extent permitted by the JHU equivalency formula.* * *JHU Equivalency Formula:* 18 graduate degree credits may substitute for one year of experience. For jobs where equivalency is permitted, up to two years of non-related college course work may be applied towards the total minimum education/experience required for the respective job. Special Knowledge, Skills & Abilities: Extraction, quantification, and handling of DNA, RNA, and protein from cells and tissues. Endpoint and quantitative PCR on genomic DNA and cDNA. Coordination and interpretation of Sanger sequence and coordination of DNA/RNA samples with Hopkins core lab for NGS. Familiarity with cell culture and mouse studies, with enthusiasm for learning to master these techniques, but extensive experience not required. Operation of centrifuge, pH meter, analytical balance, incubator, spectrophotometer, light microscope, imaging systems and laminar flow hoods. Performing established laboratory assays under the supervision of the faculty investigator. Familiarity with theory behind experiments; ability to apply analytical skills to interpret experiments using positive and negative controls. Enthusiasm to learn and master many other new techniques.
